Like EXOCET, FluoroCet measures the activity of acetecylcholinesterase (AChE), a known exosome protein, although the more sensitive FluoroCet assay relies on readout of fluorescence emission at 590-600 nm. To enable calculation of exosome abundance in your target sample, each kit includes standards calibrated to known amounts of human exosomes as determined by NanoSight analysis. How It Works

Exosome quantitation with FluoroCet is quick and easy

  1. Calibrate your fluorescence plate reader
  2. Isolate exosomes (ExoQuick, ExoQuick-TC, ExoQuick PLUS, and ExoQuick-TC PLUS are all excellent methods)
  3. Lyse exosomes with the included Exosome Lysis Buffer
  4. Measure AChE activity—add the included buffer and incubate for 20 minutes (or appropriate time as determined during the calibration step) at room temperature in the dark, and then measure fluorescence using excitation at 530–570 nm and emission at 590–600 nm.
